FBI agent, Malcolm Turner and his partner, John are assigned to capture a escaped convict known as Lester Vesco, who was originally serving a life sentence in prison but escaped his cell and killed a prison doctor and took off in his car. Malcolm and John are sent to Cartersville, Georgia to stake out the house of a elderly woman, Hattie Mae Pierce (simply known as "Big Momma") who is the grandmother of Lester's ex-girlfriend, Sherry Pierce. The film stars Martin Lawrence as an FBI agent who is tasked with tracking down an escaped convict and his loot, by going undercover as the estranged grandmother of his former girlfriend, unaware of the bond he will form with her. Surrounded by attractive young women, Trent nearly blows his cover, but befriends a girl named Haley Robinson. The headmistress announces that a historic music box has been stolen from the library, and Malcolm deduces that it contains the flash drive. While scoping out the library, Malcolm encounters security guard Kurtis Kool, who attempts to woo him while giving a tour. Seeing a picture of Kurtis with Canetti, Malcolm realizes that he is the friend, and tries to find out more about the music box.

Malcolm, in an attempt to capture Russian gang member Chirkoff, uses an informant named Canetti to deliver a flash drive to the gang. Canetti reveals that the flash drive is empty and a duplicate is hidden with a friend at the Georgia Girls School for the Arts. FBI agent Malcolm Turner is elated to learn that his stepson, Trent Pierce, has been accepted to attend Duke University.

Meanwhile, he resumes his life at home with Sherry, Trent, and their newborn baby. Although Malcolm finds evidence that Tom is working alongside a notorious hacker, Sherry tracks him down after discovering he lied to her and is appalled to discover he is working on another case posing as her grandmother. The matter is then made worse when Morales discover him interfering in her case, and allows him to assist as long as he maintains his cover and retains Kevin as his partner. After Leah threatens to fire Big Momma for her handling of her chores, Malcolm spends the night doing all of the work and successfully wins back her favor.
Trent accidentally draws their attention as he attempts to pursue Haley and explain himself. Just as he recovers the flash drive, the gang members catch up and hold him at gunpoint. Big Momma interferes and provides an escape, but all three are caught again and Malcolm's disguise is revealed. Just as Chirkoff is about to kill them, Kurtis arrives with a taser and saves the day. Although Malcolm asks to assist, his boss refuses to let him interfere in the investigation being conducted by agents Liliana Morales and Kevin Keneally.
Lester tracks Sherry down to Big Momma's house and later puts her in danger. Malcolm swiftly breaks into the house, whereupon he tackles Lester and subdues him after John is shot, but at the cost of his disguise being dropped in front of everyone, including Sherry and Trent. After Lester is arrested and the money recovered, Malcolm finds that Sherry and Trent refuse to speak to him, heartbroken by his deception.

Trent and Haley reconcile, and Malcolm signs the former’s contract, only to have him tear it up and reveal his new plan to attend college. As the film ends, Malcolm and Trent make an agreement to keep the whole ordeal a secret from Sherry. Meanwhile, the gang members approach Trent's best friends, posing as record producers, and encourage them to notify them of Trent's whereabouts. Trent sets up a date between Haley; he reverts to his true self and the date goes well, but an encounter with his best friends alerts the gang members to tail them. While the case is ended and he has made amends with Sherry, Malcolm decides to stay in California a while longer in order to assist Carrie, after helping her team improve themselves for an upcoming cheerleading championships. Helping them to successfully win, Malcolm departs, leaving behind a letter from Big Momma bidding farewell to the Fullers and hints that their paths will cross again someday.

After attending a church service with them, where he compels Sherry in his disguise to not hold any secrets, Malcolm finds the missing money in Trent's footlocker. This happens after returning home to a surprise birthday party for the real Big Momma. His discovery is witnessed by Sherry, who tells him she had no idea what Lester was planning, and did not report her key was stolen out of fear of being fired.
